Description
PE 22-28 is a seven-amino-acid sequence derived from spadin, the propeptide cleaved from neurotensin receptor-3 (sortilin).
Mechanism: Selectively inhibits the TREK-1 potassium channel, a target implicated in depression-resistance phenotypes. TREK-1 blockade enhances serotonin and dopamine signalling and promotes hippocampal neurogenesis.
Research findings: Murine forced-swim and tail-suspension models reported rapid antidepressant-like effects within hours rather than the weeks required for SSRIs. PE 22-28 is studied as a fast-acting non-monoaminergic antidepressant research reference.
Handling: Lyophilized peptide. Store sealed at -20°C for long-term stability or 2-8°C short-term. Reconstitute with bacteriostatic water or 0.6% acetic acid as appropriate to the molecule.
