Description
Semaglutide is a long-acting glucagon-like peptide-1 (GLP-1) receptor agonist with 94% sequence homology to native GLP-1 and amino acid substitutions that confer resistance to DPP-4 cleavage.
Mechanism: Activates pancreatic beta-cell GLP-1 receptors to enhance glucose-dependent insulin secretion, suppress glucagon, slow gastric emptying, and act on hypothalamic satiety centres.
Research findings: The STEP and SUSTAIN clinical programs documented HbA1c reductions of 1.5-1.8% and mean body-weight reductions of 12-15% over 68 weeks in adults with obesity. Subsequent SELECT trial data showed cardiovascular event reductions in established CVD populations.
Handling: Lyophilized peptide. Store sealed at -20°C for long-term stability or 2-8°C short-term. Reconstitute with bacteriostatic water or 0.6% acetic acid as appropriate to the molecule.
